Design and Fabrication of Biodegradable Polymer Porous Scaffolds and Biomimetic Matrices for Tissue Engineering

报告摘要:

 

 

 

Porous scaffolds of biodegradable polymers and biomimetic matrices play an important role to control cell functions and to guide new tissue formation in tissue engineering. We have developed a few types of porous scaffolds of biodegradable polymers and biomimetic matrices from cultured cells. This talk will highlight the design, preparation and application of these porous scaffolds and matrices. The porous scaffolds include funnel-like porous scaffolds and hybrid scaffolds of biodegradable synthetic polymers and naturally derived polymers. Funnel-like porous scaffolds were prepared by using ice particulates as a template and had a unique pore structure with large open pores on the top surface that communicated with interconnected inner bulk small pores. Hybrid porous scaffolds were prepared by forming collagen sponge or microsponge in the openings of porous skeletons of mechanically strong synthetic polymers. PLGA-collagen hybrid mesh, PLGA-collagen hybrid sponge and cylinder-type PLLA-collagen hybrid scaffolds were prepared by this hybridization method and used for 3D culture of chondrocytes for cartilage tissue engineering. The hybrid scaffolds combined the advantages of both types of polymers. Biomimetic matrices include stepwise tissue development-mimicking matrices and ECM scaffolds. Stepwise tissue development-mimicking matrices that mimicked the in vivo developmental ECM were prepared by decellularizing serially differentiated mesenchymal stem cells (MSCs). Stepwise osteogenesis- and adipogenesis-mimicking matrices that mimicked the ECM throughout each step of osteogenesis and adipogenesis were formed using the ECM of MSCs that underwent osteogenesis or adipogenesis in vitro. The matrices showed tissue- and stage-specific effects on differentiation of MSCs. ECM scaffolds were prepared by 3D culture of cells in a selectively removable template. The intracellular components and template were selectively removed after cell culture to obtain the ECM scaffolds. Human MSCs, dermal fibroblasts and chondrocytes were used to prepare the scaffolds. Composition of the ECM scaffolds depended on the cell type and cell phenotype used to prepare the scaffolds. Implantation of ECM scaffolds using mouse model demonstrated mild host tissue response and good biocompatibility of ECM scaffolds. These porous scaffolds and biomimetic matrices will be useful for tissue engineering and controlling stem cell differentiation.
